how to test a project in real time

Hi Everybody,
I am new to this forum.I recently got H1 .I want to try job in testing.I have read some topics on testing and got some information on testing.I just want to know on what topics I have to concentrate regarding this I am confused.Can anybody tell me what topics I have to concentrate more .I need real time implentation of manual testing considering examples like yahoo mail i.e i want to how how we conduct manual testing on yahoomail.
Can anybody guide me on this.I also want automated testing on yahoomail
using QTP,Loadrunner,Winrunner,Testdirector.
